Isolation of individual compounds using analytical HPLC and Preparative HPLC. Specifically, the activity of the participant will address one sub-objective of the project:

Sub-objective 1.3: Production of activated foods from rice for applications in human diets to promote good health: Develop methods for the induction of phytoalexins and bioactive compounds in rice for comparison against soybean and sugarcane. Completed, this research will result in food products enriched with bioactive compounds. In addition, individual bioactive compounds will be isolated from treated plant materials to determine biological activities of the components. Several analyses of plant extracts and foods will include: phytoalexin content, polyphenolic components, total phenolics, soluble and insoluble fiber, and starch digestibility.

Learning Objectives: The selected candidate will receive extensive training in the analysis of plant compounds using UPLC-MS and utilization of HPLC and Preparative HPLC. Additionally, they will receive training in the development of plant sprouts for induction of phytoalexins for production of foods. The participant will be involved in a research team that includes a plant physiologist and a sensory scientist. The participant will also collaborate with researchers at Tulane Medical School that will be conducting cell-based experiments on plant extracts and isolated compounds.

This data is used to evaluate and prioritize chemical and nonchemical stressors based on the potential for human and ecological exposure, health impacts, and ecosystem degradation.
Responsibilities

As a team member, you will conduct laboratory work, generate, and organize laboratory data, perform data analysis, and interface with experts in the respective technical specialties. The team member provide analytical laboratory support and analytical assistance to the ORD-Athens mass spectrometry laboratory in support of the Metabolomics team. The team member will be under the mentorship of CEMM researchers for training in liquid chromatography (LC) and high-resolution accurate mass (HRAM) mass spectrometry (MS) systems. The candidate will assist laboratory researchers with maintenance of the laboratory, sample processing and inventory management, routine data analysis, and some administrative support functions. The team member will maintain careful and accurate records in designated laboratory notebooks. These notebooks and all other data produced under this order will be the property of the Environmental Protection Agency.

How you will apply your skills

Analytical and Laboratory support tasks will include:
• Maintaining, managing, and organizing laboratory facilities, samples, supplies, and reagents
• Preparation of standard solvents and solutions
• Routine instrument calibrations
• Receiving and logging of biological/environmental samples
• Processing of received samples for LC-MS analysis
• Conducting experiments using liquid chromatography and HRAM-MS methods. Experiments will be planned by consulting with research investigators regarding sample preparation, analyte detection and (semi-)quantification, and data quality control methods
• Maintaining records and documentation associated with the conduct of these experiments, in accordance with Quality Assurance plans

Communications-related responsibilities will include:
• Participating as a member of a multi-disciplinary research team
• Presenting data summaries and results to EPA team members
• Assisting in the preparation of analysis summary documents such as manuscripts, presentations, fact sheets, and reports.
• Documenting daily laboratory activities in designated laboratory notebooks
• Presenting work performed at a scientific conference as required.

Required Knowledge, Skills, Work Experience, and Education
• At least one year's experience in graduate school performing (semi-)quantitative chemical analysis including at least 16 hours of laboratory procedures such as pipetting, sample extractions, chromatography, and/or mass spectrometry;
• Working knowledge of physiochemical separation by liquid chromatography and detection by HRAM mass spectrometry;
• Prior experience working with environmental and/or biological samples;
• Experience using at least one of the following: R, Python, and/or Microsoft Excel, to summarize and manipulate data files, manage data, prepare graphics, and conduct basic data analysis such as summary statistics;
• Strong written, oral, and electronic communication skills.
• If still enrolled in school a transcript will be required each semester to show applicant in good standing with university.

Responsibilities

This project is developing the Human Exposure Model (HEM) to assess exposures to chemicals from the use of consumer products. The team member will assist with developing datasets and decision rules for various modules of HEM, and assist in the testing or evaluation of the HEM model and data bases of product composition information. The work of the team member may include gathering additional datasets from online sources, formatting datasets into standard templates and uploading into databases, applying out-of-the-box thinking to determine rules to parametrize models of human behavior and product usage, and testing and evaluating ease-of-use of software.

• Manipulation of information on chemicals stored in spreadsheets. Collecting, curating, and organizing data (e.g. in databases) on composition of consumer products (chemical names and related information).
• Assist in the development of scientific modules that will be part of the HEM (e.g. modeling occupational exposure to chemicals, end-of-product-life exposures, etc.). This could also include developing user tools, such as a graphical user interface (GUI) for the HEM interface.

Communications-related responsibilities will include:
• Participate as a member of a multi-disciplinary research team;
• Interact with other members of the development team as well as EPA scientists;
• Document database development efforts; and
• May be asked to present work performed as a poster at a scientific conference.

Required Knowledge, Skills, Work Experience, and Education
• General understanding of quantitative techniques, basic statistics, and use of spreadsheets;
• Strong reading comprehension skills and experience logically interpreting pieces of information from a variety of data source types;
• Experience integrating partial pieces of information and applying out-of-the box thinking to determine how to use limited available data to fill data gaps; and
• Experience with any computer programming language.

The Watershed & Ecosystem Characterization Division (WECD) conducts research to advance EPA’s ability to characterize the presence, transport, transformation, sources, and impacts of contaminants in watersheds and ecological systems. CEMM/WECD scientists:
• Develop methods to measure chemical and microbial pollutants in a variety of environmental media (soil, dust, water, and other biota) to support implementation of the Toxic Substances Control Act (TSCA), the Comprehensive Environmental Response, Compensation, and Liability Act (CERCLA, also known as Superfund), the Resource Conservation and Recovery Act (RCRA), Clean Water Act (CWA), Safe Drinking Water Act (SDWA), the Harmful Algal Bloom and Hypoxia Research and Control Act, the Beach Act and other environmental statues.
• Conduct ecological monitoring of chemical and biological stressors at multiple scales to characterize the condition of environmental systems through the development and application of novel field indicators, environmental genomics, and geospatial tools.
• Develops, evaluates, and applies watershed management tools to characterize both ecological response and economic benefits. The methods, tools, and technologies inform effective watershed management practices as well as minimize health risks to various chemical and microbial contaminants.

As a team member, you will assist EPA staff with the administrative duties required to support and maintain day-to-day program operations, including the administration, management, and logistics of government resources.
